Objective: Notch/Delta signal is the conservative way with regulation function in embryonic development and expresses in many vertebrates and invertebrates. This signal possesses decisive role in cell differentiation, fate choose and development. Recently more researchs of Notch signal in the development of nerve stem cells have been studyed, but the research of Notch signal in the development of mammary stem cells is rare. Stem cell biology has become a hot field in the study of cytobioogy. People begin to realize that the tumour is a kind of stem cells disease. As regrds the disease of mammary carcinoma,some research had proved that the existence of the stem cell in breast epithelia.A group of researchers have utilized immunostaining to show the different phenotypic characteristic of breast epithelial stem (progentior)cell, differentiated cell and intermediate cell. In our research, we have replicated animal modle of breast hyperplastic lesion in rats and utilized immunohistochemical technique and RT-PCR to detect the expressesion of proteinum and mRNA in these modles. We also collected the mammary tissue of operation and detect the expression of CK5, CK8 and P63 in 10 case of nomal mammary tissues, 20 case of benign ductal hyperplasic, 14 case of atypical hyperplasia, 6 case of ductal cancer in situ and 48 case of invasive ductal cancer, in order to investigate the role of Notch1 signal in the differention and development of human and rat mammary stem cells, and reveal the rule and mechanism of...
