| Background:Oral leukoplakia(OLK)is the commonest precancerous disease in oral cavity, and its canceration rate reach 3%-5%. The average course of this disease reach to 8.2 years. More attention is paid to OLK now because of its higher rate of canceration and longer process of canceration. A few of OLK will finally turn into squamous cell carcinoma of oral(OSCC), and OSCC is the most common malignant tumor in the oral cavity ,accounting for more than 80% of the oral maligant tumor .Therefore, Detecting OLK's carcerating potential and making early correct diagnoses on OSCC have important directive significance to clinical works.Study show that COX-2 is overexpressed in a variety of precancerous or malignant lesions inclouding OLK and Head and Neck Squamous Cell Carcinoma (HNSCC). Upregulated levels of COX-2 may contribute to carcinogenesis by modulating renobiotic metabolism, apoptosis , immune surveillance , invasion, metastasis and angiogenesis .Bcl-2 gene was an important anti-apoptotic gene and its product had been demonstrated to prevent apoptosis. Ki-67 was considered the best index of cell proliferating activity. At present, few study on the expression of Cox-2 , Bcl-2 and Ki-67 in OLK and OSCC is known.
