Objective To observe the effects of pioglitazone and tumor necrosis factor- α on adiponectin mRNA expression and secretion of adiponectin protein in 3T3-L1 adipocytes. Methods Treat the 3T3-L1 adipocytes with pioglitazone and TNF- a under diversity concentration and diversity phase, using the RT-PCR to measure the adiponectin mRNA expression level, and using the ELISA to measure secretion of adiponectin protein in cell culture fluid. Results (1) Adiponectin mRNA and adiponectin protein were not detected in preadipocyte. With the differentiation from 3T3-L1 preadipocytes to 3T3-L1 adipocytes, the expression level of was increased accordingly. (2) 0ng/ml,0.1ng/ml, 1ng/ml, 10ng/ml, 100 ng/ml of TNF-α treated in 3T3-L1 adipocyte for 48 hours, the relative adiponectin mRNA values were decreased in a dose-dependent manner (1ng/ml-12. 6%, 10ng/ml-29. 3%, 100 ng/ml- 68. 65% vs ccontrol ) .100 ng/ml of TNF- α treated 3T3-L1 adipocyte of differentiation, the adiponectin mRNA was not detected for 2 days, but the adiponectin mRNA decreased in a time-dependent manner (4days-3.40%, 6days-10. 54%, 8days-17. 60% vs ccontrol). 100 ng/ml of TNF-α treated in 3T3-L1 adipocyte , the adiponectin mRNA decreased in a time-dependent manner (24 hours - 12. 38%, 48 hours - 63. 44%, 72 hours - 64. 33% vs ccontrol). The concentration of adiponectin in the cell culture fluid were depressed by TNF-a in 3T3-L1 adipocyte in a dose- dependent manner.
