Introduction Lung cancer is the most commonly fatal malignancy, it causes the greatest number of cancer death worldwide for all forms of cancer. Successful treatment of this diease depends on its early detection,but clinical detection of lung cancer usually occurs late in the disease.It have reported that a monoclonal antibody, 703D4, which recognizes heterogeneous nuclear ribonucleoprotein A2/B1 ( hnRNPA2/B1 ) can frequently detect lung cancer in exfoliated sputum epithelial cells 1-2 year earlier than routine X-ray or sputum cytomorphology. Heterogeneous nuclear ribonucleoprotein A2/Blis a kind of embryo antigen correlated to the initiation and the development of lung cancer. It has been recently reported that hnRNPA2/B1 pooly expressed in normal cells and overexpressed in the beginning of lung cancer. The level of hnRNPA2/B1 raised in the developing process of lung cancer. After reaching a peak value it began lower little. This suggests that hnRNPA2/B1 is an early marker of lung epithelial transformation and carcinogenesis.Objective: Clone the hnRNPA2/Bl gene and construct the recombinant prokaryotic plasmid to induce expression of hnRNPA2/B1 fusion protein.Methods: The cDNA of hnRNPA2/B1 gene was cloned from the total RNA of human muscle cell by RT-PCR. After sequencing, the gene was cloned into the expression vector PGEX-4T-1 to construct recombinant plasmid PGEX-A2/B1. The BL21 contained expression plasmid wsa induced by IPTG.Result: The full-length cDNA of hnRNPA2/Bl gene was cloned and confirmed by DNA sequencing. The prokaryotic expression vector for the hnRNPA2/B1 was constructed, and the fusion protein was expressed in E.coli after the IPTG induction.
