| Objectives: ①Tb culture the rats' bone marrow-derived mesenchymal stem cells (MSCs) in vitro, observe the biological features of MSCs. ②Differentiate MSCs into neural-like cells with homogenate of damaged brain tissue. ③Investigate the effect of (MSCs) administered by the way of internal carotid artery on functional outcome after traumatic brain injury in adult rats. ④ Observe the influence of anti-oxidation and iNOS expression after transplantation.Methods: ①MSCs were isolated from the bone marrow of SD rats, cultured in vitro and observed their biological features. ②We induced MSCs into neural-like cells with the homogenate of damaged brain tissue, evaluated them with immunocytochemistry. ③The adult rats undergone traumatic brain injury (TBI), MSCs that marked by BrdU were transplanted from the internal carotid artery. ④We evaluated the neurological function of rats by a normal method—neurological severity score and the expression of BrdU with immunohistochemistry after transplantation. ⑤In addition, not only the content of SOD, NOS, OH~- in the experimental group rats brain tissue have been detected, but also the contrast group rats. The two group rats' expression of iNOS also have been detected. Analyses of statistics have been use for find the difference of the two groups.Results: ①Rat MSCs showed active proliferative ability in passage culture and can expand rapidly in adherent cultures.MSCs can Pure by transfer of culture, after 20 times transfer, MSCs also can growth very well, and their biological features were stable. ②... |
