Prostate cancer(PC) occurs the most commonly in urinary system of man.This ranks prostate cancer second only to lung cancer as a cause of cancer-related death among men in American. In recent years ,with the change of population aging and food structure,the morbidity and mortality of prostate cancer in our country have the trend to increase year by year.The early metastasis of prostate cancer is the main reason of the failure treatment. Gene expression related to the regulation of cell cycle,apoptosis,adherency,activity and angiogenesis changes notably when prostate cancer metastasize.And the gene expression interact as both cause and effect with disease progress.It needs to establish the corresponding animal models and methods to understand the malignant biological behavior.In this study,we will establish the metastatic models of human prostate cancer cell to represent clinical-cancer better,especially with regard to experimental study by optimizing the human prostate cancer cell line PC3,which lays the foundation of deep studing the mechanisms of human prostate cancer metastasis,looking for the new markers of prostate cancer metastasis and developing effective prevention and intervention strategies to manage and treat this disease.So we carried out the following three-part work.1. Isolation and establishment of human prostate cancer cell clones with different metastatic potential.The single cell that isolated from the synchronized human prostate cancer cell PC3 using limiting dilution method was cultured in vitro.When the single cell grow into cell clone,it was inoculated into the nude mice.The two cell clones with the biggest different metastatic potential were isolated from the becoming fifteen single cell clones by inoculating them into the nude mice. Then the two target cell clones were verified by enlarging the cell culture and increasing the amount of nude mice.Thus human prostate cancer cell clones with high(PC3-H) and low metastatic...
