Objective The spread of epileptic activity in brain is not random, which generated and spread by specific anatomic pathways such as substantia nigra. Substantia nigra is an important nucleus in extrapyramidal system, which not only takes part in the motion adjustment, but plays a role in the adjustment of epilepsy. The previous study shows the substantia nigra plays a "gating" role in the epileptic activity. Substantia nigra can inhibit some kinds of seizures by inhibiting the epileptic activity through GABAergic neurons. To study the changes of GABAergic neurons in substantia nigra of the epileptic model induced by pilocarpine in rat.Methods Seventy adult male Wistar rats were randomly divided into group control and pilocarpine group. Group control was injected in the abdominal cavity of the rats,and pilocarpine group was performed in pilocarpin group.Rats were killed respectly at 1 hour, 6 hours, 12hours, 24hours and 72 hours , after treatment. GABAergic neuron section in substantia nigra was estimated by immunofluorescence and semi-quantitated according to marked cells.Results The positive cells showed green fluor, were mainly located substantia nigra pars reticularis. The number of GABAergic neurons in substantia nigra reduced along with epilepsy paroxysm at 1 hour, compared with that in group control.
