To study the expression and significance of adrenomedullin in placenta of preeclampsia, as well as the possible pathological and physiological role of adrenomedullin in preeclampsia and eclampsia.2. To study the relationship between ADM concentration in maternal peripheral vein blood and preeclampsia and eclampsia. And investigate the relationship with Severity of the cases.3. Analyze the relationship between ADM concentration and clinical parameter, such as MAP, neonate birth weight and placental weight.MethodsThe expression of adrenomedullin in the placenta of 19 cases of normal third-trimester pregnant women, 40 cases of preeclampsia (18 mild and 22 severe)and 5 cases of eclampsia were detected by Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR)and immunohistochemistry test(SP method). Moreover, We assay ADM on serum of maternal peripheral blood using enzyme immunoassay(EIA).ResultsWe detected ADM mRNA and ADM peptide in both placenta of control pregnant women and cases; ADM was mainly detected in chorion, decidua baseplate, syneytiotrophoblast and so on. Meanwhile We found that ADM mRNA expression in the placenta obtained from women with preeclampsia or eclampsia was increased compared with that in normotensive controls(1.0966±0.1533vs0.8979±0.0520) (P<0.01).
