Semen Cassiae is the dried ripe seed of Leguminosae plant Cassia obtusifolia L. or Cassia tora L, which is being recorded in China pharmacopoeia. The study screen out the active compound from Semen Cassiae in vivo and vitro evaluation, and prove it is the specific component in Semen Cassia. Basing these researches, we study the preparation method of aurantio-obtusin and the quality specification of Semen Cassiae.The research studied the effective components of reducing blood lipid in Semen Cassiae, and Six compounds had been isolated and identified as obtusifolin, aurantio-obtusin, Gluco-obtusin, Gluco-obtusifolin, Daidzin and Cassitoroside.Develop the LDLR reporter gene system to research the action of reducing blood lipid of the six compounds. Result shows aurantio-obtusin could enhance the gene expression of LDLR and could reduce blood lipid.And it is proved by hyperlipidemia model by SD mice.Aurantio-obtusin is indicated to be the specific component, in Semen Cassiae by the study of thin layer chromatography and high performance liquid chromatography.We setup the quality specification of Semen Cassiae. Test water, heavy metals and As, total ash and acid-insoluble ash, ethanol-soluble extraction. We take aurantio-obtusin as reference substance to setup the TLC method of Semen Cassiae, and aurantio-obtusin are tested in 16 samples. And setup the RP-HPLC method of Semen Cassiae. The method is convenient and sensitive. We determined sixteen type of Semen Cassiae sample from different habitat by using this method. Then we suggest the content of aurantio-obtusin in Semen Cassiae should be above 0.350mg/g.We studied RP-HPLC fingerprint of 16 samples of Semen Cassiae. The result shows this method is stable, reliable and has good recurrence. And there are 13 batches sample have the similar common-characteristic in the fingerprint...
