OBJECTIVE: Methotrexate is a major component of current treatment regimens for children with acute lymphocytic leukemia (ALL). But its high individual differences require to determine its concentration in serum and to monitor its concentration in children with ALL. At present, there are many methods to determine its concentration, such as FPIA, HPLC, UV,et al.Our objective is to develop a HPLC method to determine the methotrexate(MTX) concentration in plasma and to monitor its concentration in children with acute lymphocytic leukemia (ALL) treated by MTX and CF (HD-MTX-CF). The study provides the basis of the treating and the saving in clinic.METHODS: The MTX concentration in plasma was monitored by the LC-10ATvp HPLC system at 302 nm. The mobile phase consisted of 0.3% phosphoric acid and acetonitrile(85:15).
