Objective:To establish a novel real-time fluorescent PCR method to detect HCV with high sensitivity, specificity and stability using the duplex scorpion probe.Method:The recombinant vector pMD18-T-HCV-5'-NCR was used as the standard template. The duplex scorpion probe was designed according to the cloned gene sequence. And then the PCR reaction system was optimized and evaluatedResults: 1) The recombinant vector was cloned successfully; 2) Methodology analysis showed the duplex scorpion probe detecting system was established with large detecting range, high sensitivity, specificity, stability and accuracy. The difference between this method and the UV is insignificant; 3) After applied in clinical experiments, this novel fluorescent quantitative method has more advantages than the DA'AN HCV detecting kit.Conclusion: A novel real time fluorescence quantitative PCR detecting method for HCV has been developed, which would lay a substantial foundation for exploring new clinical diagnostic kits using the...
