| Objective: To explore the effect of different doses of cyclophos- phamide (CTX) on drug-resistance of implanted human A549 lung cancer in nude mice, and investigate the antitumor effect and toxicity, in order to propose a new regimen of chemotherapy and provide theoretical evidence of its clinical application. Methods: BALB/C nude mice bearing A549 lung carcinoma were randomized into several groups, which received low-dose metronomic (LDM) CTX,maximum tolerate dose(MTD) CTX therapy respectively, then we investigate changes of tumor volume, weight and toxicity. At the end of experiment , tumors were resected for immunohistochemical staining . Expression of glutathione S-transferase-Ï€(GST-Ï€) and lung resistance-related protein(LRP) were detected by immunohistochemistry SP method using monoclonal antibody to their protein. Results:â‘´In LDM CTX therapeutic group tumor grew fast at first , then it slow down gradually and grew slowly for a long time, while in... |
