1. Determination of trans-resveratrol(t-Res) and trans-piceid(t-PD) in rat plasma by HPLC and their pharmacokineticsObjective: To establish methods for the determination of t-Res and t-PD in rat plasma by HPLC. And to investigate and compare with the pharmacokinetic parameters of t-Res and t-PD in rats.Methods: The Sprague-Dawley(SD) rats were divided into 3 groups randomly, sixty in each group. Each group was taken with different oral doses of t-Res (29.2, 58.5, 116.9mgkg-1 ). Blood samples were collected from eye sockets at the setting time (0.083, 0.167, 0.25, 0.333, 0.417, 0.5, 0.667, 0.833, 1, 1.5, 2, 2.5h after administration) and drawn into tubes with heparin sodium. Plasma was obtained by centrifugation at 3,000 rpm for 10 min. Equal volume of acetonitrile was added, which was mixed to precipitate the plasma protein, then added double volume of distilled water and centrifuged at 12,000 rpm for 10 min. The supernatant (20μl) was injected into the HPLC system. The analytical column was Hypersil BDS C18 column (200mmx4.0mm, 5μm). The mobile phase was consisted of acetonitrile-1% glacial acetic acid (25:75). The flow rate was 1.0 ml·min-1 . The UV detection wavelength was set at 306nm.
