The Research Of Smedds Capsule Of Nimodipine

Nimodipine (NMD) is a dihydropyridine calcium channel blocker which is very effective to cerebral vessels. Appropriate dose of NMD can expand selectively cerebral vessels, almost have no effect to peripheral vessels. Nimodipine, entering the cerebral tissue, combines reversiblly and specially with the recipient of calcium channel. And it also regulates the calcium to flow into nerve cells and influences the electrical properties of nerve cells and the balance of nerve medium.Nimodipine is good to cure aged Dysencephalia and at present the most-widely used drug to aged Dementia. Nimodipine is a kind of drug with low water solubility,low dissolution and low bioavailability, Especially to tablets of NMD made in China. So we study and make the self-microemulsifying drug delivery syestem(SMEDDS) of NMD,which can markedly improve the dissolution and bioavailability of nimodipine.We investigated the saturated solubilities of NMD in various vehicles and the coeffect between all kinds of oils and surfactants. According the results, we decided to design the formulae of SMEDDS from two point of views. One of SMEDDS formulae is composed of cosurfactant,surfactant,oil and another includes mixed surfactants and oil. We respectively studied the pseudo-ternary phase diagrams of two kinds of SMEDDS, the result of droplet size s after self-microemulsificaion ,the easy level of self-microemulsification.
