Poliomyelitis is one kind of infectious viral diseases that spread wide and invade central nervous system. Great success has been gained depending on oral poliovirus vaccine (OPV) since launching the campaign of elimination poliomyelitis in 1988. But OPV may generate vaccine correlation paralysis and virus-derived poliovirus. In contrast, inactivated poliovirus vaccine (IPV) can overcome above mentioned shortcomings. WHO points out that inactivated poliovirus vaccine is needed to maintain immune level to poliomyelitis after the realization of goal of eradicating poliomyelitis. More and more importance has been attached on Sabin inactivated poliovirus vaccine (Sabin IPV) because of its safety. Institute of Medical Biology developed Sabin IPV. In order to study detect the D titer of IPV , Monoclonal antibodies against D-antigen of type Ⅱ Sabin strain were prepared by hybridoma technique. Indirect ELISA and micro neutralization analysis were used to identify its specificity and titer. Four hybridomas secreting monoclonal antibody(McAb) against the D-antigen of poliovirus type Ⅱ Sabin strain were selected out. The ascetic antibody titers measured with ELISA and neutralization assay were 1: 2.4 × 10~6, 1: 1.3 ×10~4 respectively. The McAb only reacted with the D-antigen of poliovirus type Ⅱ Sabin strain, but not with C-antigen of poliovirus type Ⅱ Sabin strain. The potency of the D-antigen Ⅱ -type in sabin trivalent inactivated poliovirus vaccine was determined, by enzyme-linked immunosorbent assay using monoclonal antibody and multiantibody simultaneously. In the range of 0. 25Du/ml-2 Du/ml, their relation coefficients are 0.998, 0.966, 0.903; 1.000, 0.998, 0.986 respectively. The McAb can be applied in the standardization of determining the potency in vitro.
