ObjectiveTo investigate the level of antibiotic resistance, the frequency of class I integrons in Escherichia coli isolated from healthy human in Shenzhen, and analyze the relationship between resistance genes carried by class I integrons and antibiotic resistance phenotypes. To observe the role of class I integrons in the acquisition and transmission of antibiotic resistance among Gram-negative bacilli. To study the molecular mechanism of integrons and gene cassettes in mediation and transmission of antibiotic resistance. The study laid a theoretical foundation for the control of antibiotic resistance and multi-drug resistant bacteria, the reasonable usage of antibiotics in clinical treatment. MthodsEscherichia coli isolated from healthy human were identified. Susceptibility of 150 Escherichia coli islates to 16 different antibiotics was determined by the disk diffusion method. Analyze the plasmid profile of isolates and the resistance strains were classified. The presence of class I integrase(intI1) in the isolates were detected by PCR amplifing total DNA. Class I integrons of intI1 positive strains were detected by PCR amplifing total DNA and plasmid DNA respectively. The amplicons of class I integrons with identical length were analyzed by RFLP and class I integrons with different restriction patterns were sequenced.
