| Objective: To investigate the damage of ischemia-like in the primary cultured rat's cortical neurons .To explore protective effects of aspirin on the neurons and the optimal dosage.Methods: Cortical neurons cultured for 7 days were randomly divided into five groups: ischemia-like control group, ischemic preconditioning group and plus different concentrations of aspirin preconditioning groups. Then morphology of neurons was observed under inverted microscope. The survival rates of neurons at different groups were analyzed with MTT reduction assay. The apoptosis of neurons was detected with the method of and the apoptotic rate of neurons was measured using flow cytometry.Results: There was a significant difference between the ischemia-like group and the lower aspirin preconditioning group (P<0.01). The lower concentration aspirin preconditioning could reduce apoptosis of neuron and increase survival rates of neuron. But there was no difference between ischemia-like control group and the medium, high concentration aspirin groups (P>0.05) .Conclusion: Using the lower concentration aspirin before ischemia-like can alleviates the toxicity damage of neurons caused by ischemia-like through reducing neuron apoptosis, and increase the survival rate of neurons which suggests that aspirin is an effective neuroprotection . |
