Experimental Study On The Treatment By Mesenchymal Stem Cells To Emphysema Animal Model

Objective: The aim of this study was to culture the mesenchymal stem cells (MSCs) in vitro and inject them into the tail vein of the emphysema model rat. Four weeks later, we detected the pathological effects to the emphysema lung tissue. Maybe we can find a new and effective treatment to emphysema.Methods: Wistar rats with initial weight of 250±10g were used in this study .Animals were randomly divided into four groups. Normal group(A),model group(B),treatment group(C) and control group(D). B,C,D group rats received a single intratracheal instillation of pancreatic porcine elastase (PPE). One mouth later, the emphysema models were completed. MSCs were cultured in vitro and labeled with BrdU. MSCs were injected into tail vein of the D group rats. Four weeks after the injection, artery blood gas was analysised. Lung tissue pathological changes were observed and recorded the number of alveoli (Na), and calculated the mean number of alveoli (MNa) and mean area of alveoli (MAA). The same time, MSCs were examined by immuocytochemical method.Result: There were no significant differences in artery blood gas between all groups (P>0.05).Compared with rats of group A, there were significant differences in rats of group B,C and D in MNa (P<0.001). The MNa of group D was significantly more than the group B(P<0.001)and group C(P=0.02).There were no significant differences in rats of group A and D in MAA(P=0.128), and the MAA of group D was significantly smaller than the group B (P<0.001) and C (P=0.015).Conclusion: By injecting MSCs into the veins to treat experimental emphysema, the MNa of the treatment group was few than the normal group, but it was more than the model and control groups. It indicated that MSCs was effective to the experimental emphysema treatment.
