Association Of T182C And G1287A Polymorphism In Norepinephrine Transporter Gene And Major Depression Under The Controling Of Social Psychic Factors

Objective:1. To validate the influence of social psychic factors during the association study between norepinephrine transporter (NET) gene polymorphism and major depression (MD).2. To investigate the relationship between NET gene polymorphism (including T182C and G1287A) and morbility, severity, suicidal idea and effect of antidepressant of MD under the controlling of social psychic factors.Methods:A case-control sample, 154 case were diagnosed by DSM-IV, 154control group were selected to match age,gender with case group. EPQ, LES, SSS and CSQ were used to evaluate the social psychic factors, such as personality, life events, social supports and coping styles. Pyrosequencing PSCE96MA was used to detect the T182Cand G1287A genetic polymorphism of the NET gene.The genotype and allele frequency distribution were compared withχ2 analysis,multivariate logistic regression was used to control social psychic factors and compute odds ratio.Results:1. After controlling the social psychic factors, the OR values of the two polymorphism were all proceed some change, which hint that social psychic factors influenced relevance between NET gene polymorphism and MD.2. Results of relationship between T182C gene polymorphism and MD.①The frequency of T allele and TT genotype in control was higher than MD group , the frequency of C allele and CC genotype in MD group was higher than control . The OR value of C allele and CC genotype was 1.692(P=0.066)and 6.232(P=0.019).②Grouped by HAMD score of 24,the OR value of TC genotype of different group was 0.42(7P=0.073),which hint TC genotype may be a protective factor of MD severity.③There were no significant differences of the distribution of NET genotypes and alleles between suicidal idea group and non-suicidal idea group, and the OR values were not get statistical significance yet.④After drug treatment of 2 weeks and 6 weeks by fluoxetine, there were no significant differences of the distribution of NET genotypes and alleles between the ineffective and effective groups, in addition the OR values were not get statistical significance yet.3.
