3,4',5-trihydroxylstilbene-3-O-β-D-glycoside (polydatin or piceid), a phenolic phytoalexin, is one of the main bioactive components in the rhizome of Polygonum cuspidatum Sieb. et Zucc. Various potential pharmacological effects have been observed both in vitro and in vivo for polydatin, including cardioprotective, anticarcinogenic, antioxidative, phytoestrogenic effects, etc. Therefore, the evaluation of the absorption, distribution, metabolism and excretion characteristics of this potentially important medical constituent is a major prerequisite for the interpretation of the pharmacological testing and the use of clinical drug aright guide. In this study we compared the pharmacokinetics characteristics of polydatin in three different forms after oral administration to rats with reversed-phase HPLC. We also investigated the tissue distribution, metabolism and excretion characteristics. Objective: To develop a method to separate extractions from Rhizoma polygoni cuspidati which meets the requirements of pharmacokinetic study; To establish a reversed-phase HPLC method for determination of polydatin in plasma and tissue samples; To study the characteristics of...
