Catarct is a kind of common disease which can cause blindness in theworld. It occurs when proteins and lipids of crystalline lens are injured byoxidization. As we expected, the peroxidase simulacrum can become a validdrug for preventing and curing catarct because it may enhance theinoxidization activity of crystalline lens, may inhibits and cuts off theoccurrence of oxidization injury. With the mother plate of microperoxidaseMP-9, DhHP-6 compound containing a histidine residue was designed to be aperoxidase simulacrum on the grounds of the structure of ascorbic acidperoxidase with a haemachrome agon. The simulacrum was developed to bean activity anti-catarct drug. The target compound shows a certain strongcatalytic activity (2.63×103U/μmoL) by enzyme activity assay in vitro. It canremove effectively oxidization injury of panniculus adiposus which wascaused by H2O2 and lipid-peroxide. It can penetrate crystalline lens membranewith a low molecular messes (<3000Da). In addition, because of havingiso-peptide linkage in molecules, the target compound have other features suchas non-sensitive to hydrolase, higher stabilization et al.. A process route standard for DhHP-6 was determined under our researchwork. Purity of DhHP-6 is up to 98% and recovery of it reaches above 30%synthesized by the technology. Anti-catarct activity of DhHP-6 was confirmedby vivo and vitro experiments. It is testified that cataract occurrence ofexperimental animal patterns is prevented effectively by DhHP-6 (15μg/mL)in low concentration.
