Objective:To observe the protective effects of acupuncture treatment on optic nerve of the rabbits model whose intraocular pressure was reduced to normal after chronic intraocular pressure elevated (IOP), and study the mechanism of it.Methods:Elevated intraocular pressure was gotten by injection of 0.3 carbomer into anterior chamber of albino rabbit. IOP was reduced to normal through conventional glaucomatous trabecuietomy after the intraocular pressure continuously for 1 months. Acupuncture treatment was used to treat the rabbits everyday in the treated group after the trabecuietomy for 4 weeks. The levels of the glutamic acid and NO in retina was measured after treatment, and the pathologic changes were observed. A computer image analysis system was used to count the retinal ganglion cells(RGCs) and the optic nerve axons . To observe the affection on the expression level of Bcl- xl and BDNF in retinal too.Results:The glutamic acid and NO levels have significantly difference between each group (P < 0.01), and the treatment group was significantly lower than those in experimental group (P<0.05,P< 0.01). Electron microscope result indicated that compared with experimental group optic nerve axons swelled relatively gentler, and the degree of damaged retina ganglion cells in treatment group was lesser than that in experimental group. The number of RGCs and optic nerve axons of treatment group was more than the experimental group (P< 0.05,P< 0.01).Expression level of Bcl- xl and BDNF in retinal was enhanced in treated group .Conclusion:Acupuncture treatment can protect the optic nerve from elevated IOP, by alleviated the damages of the retinal and optic nerve axons ultrastructure induced by chronic ocular hypertension preventing the apoptosis of RGCs, accelerate NO and glutaminic acid remove in rabbits retina and enhanced the expression level of Bel- xl and BDNF in retina... |
