| Object: To evaluate expression of TGF-β1 protein in liver that induced by vena mesenteric injection of pingyangmycin and adriamycin to investigate the mechanism of pingyangmycin in the treatment of angiocavernous. Methods: (1) Preparation of animal model: 54 rats were randomly divided into four groups. Group A (15 rats) were induced by vena mesenteric injection of pingyangmycin, Group B (15 rats) were induced by vena mesenteric injection of pingyangmycin and dexameth, Group C (15 rats) were induced by vena mesenteric injection of adriamycin, the other 9 rats (group D) are normal control group which were induced by vena mesenteric injection of normal sodium. All operation was executed under general anesthesia.(2) Obtaining specimen : Livers of 5 rats of the group A,B,C and 3 rats of normal control group were obtained on the 7th,14th and 28th day respectively after the operation. The specimens were fixed by formaldehyde solution.(3) Pathological observation: Specimens fixed by formaldehyde solution were embedded by paraffin and the thickness of continuous sections was 4 um. Haematine-eosin (HE) and immunohistochemistry stain of transforming growth factor β1 (TGF-β1) were performed.(4) The livers of rats were studied by SABC immunohistochemistry with the TGF-β1 rabbit anti-human polyclonal antibody. The intensity of staining was scored in each specimen on a scale of 0 to 3 (0=negative,... |
