Benzene is a kind of widely used industrial material and organic solvent. It is an established human carcinogen (IARC, 1982). Long-term exposure to benzene can result in lymphohematopoietic malignancies and chronic benzene poisoning. Chronic benzene poisoning is still the commonest occupational poisoning in China. If we can find a biomarker, which can not only response cumulative doses of benzene, but also prove the susceptivity of individuals to benzene, then the cancer risk of benzene can be well estimated. In addition, it is proved that the primary toxic effect of benzene is genetic damage. Study on the genetic damage of chronic benzene poisoning is helpful to protect the benzene-exposed workers' health and to prevent and cure chronic benzene poisoning.In this dissertation, the difference of GPA mutation frequency of peripheral lymphocytes was primarily investigated between the chronic benzene poisonings and benzene-exposed workers. And we also analyze factors correlated with GPA mutation frequency. In addition, the relationship between GPA mutation frequency and benzene exposure was also explored. Also we have collected peripheral blood to investigate on individual genetic susceptibility by using the single cell gel electrophoresis assay and cytokinesis-block micronuclei and 3AB index test. In short, we hope it would provide theoretic information for the hazard assessment of benzene and prevention and treatment of the chromic benzene poisoning. And we investigate further on the accuracy of GPA mutation frequency as a forecasting index for the bio-dosimeter and cancer risk.The followings are the primary results of the study:We measured GPA mutation frequency in peripheral erythrocytes from workers exposed to benzene.
