| Kiko-to is composed by Platycodin and Glycyrrhiza and is the prime recipe for throat inflammatory diseases.The thesis discusses the mechanisms of the traditional drug pair in in-vivo, enzymic, cellular levels.In the same time, comparing and evaluating the drug interaction methods in the Chinese traditional "Compound" research, the research results are listed:1 In the ear edema model, saponins of platycodin and glycyrrhiza could obviously inhibit the extent of ear edema. By the modifid isobologram method, in the dose scope of 202.5mg/kg and 135mg/kg, the interaction of the two drugs displayed to be synergism.While in the other hand,in the "removing phlegm effects"model , the two drugs only interact with each other additively.2 To further discuss the mechanisms of anti-inflammatory effect of kiko-to, we carry on the experiment of the effects of glycyrrhiza and platycodin play on the secretion of TNF-α and IL-1, although platycodon has no obvious dose-effects relationships on these two cytokines.In the experiment on TNF-α, the two drugs display synergism. This result maybe can help to explain the synergism mechanism of anti-inflammatory effects of these two drugs.In the same time, the article carry on research on the two herbs' effects on the secretion of NO in resting RAW264.7 cells and find they can enhance the secretion of NO and have rather good dose-effect relations.By the method of modified isobologram, the two drugs have obvious synergism interaction.The article also applicates drug interaction methods of isobologram and modified isobologram into the study of the two drugs' effect on the enzyme of tyrosinase and draw a conclusion that the total dose and combination ratio would affect the drug interaction effects.The article also evaluates the drug interaction method like isobologram, modified isobologram, median effect method, ANOVA, Q value method, and applicate them into the study of the mechanism of the combination usage of platycodin and licorice, the results are... |
