It is very important for the vascularization of the bone grafts in the remodeling and reparation of bone defects. The factors of angiogenesis of bone are very complicating, including cell, extracellular matrix,cytokine , biomechanics and so on . The use of cytokine ,such as VEGF, bFGF, is one of the main methods of the bone vascularization. But the high price of cytokine, and potential biosafety (Vascular neoplasms), another thorny problem, prevents it's generalization and application in clinical works. So the investigation of vascular endothelial cells to promote the bone angiogenesis maybe the new strategy .The absence and the dedifferentiation of the autoallergic endothelial cells in vitro impel researchers to look for other methods: inducing stem cells to differentiate into ECs. BM-MSCs with multi-differentiate potential ability can be induced to vascular endothelial cells by the action of cytokines of VEGF,bFGF,IGF-1,ECGS alone or jointly in vitro. So this research investigate whether fluid shear stress (FSS),like VEGF, can promote the rabbit BM-MSCs to differentiate into vascular endothelial cells.Objects:Improve the parallel flow chamber device in order to better imitate hemodynamic shear stress action on the endothelial cells; based on the device , investigate whether fluid shear stress can promote the rabbit BM-MSCs differentiation into vascular endothelial cells alone or jointly with the cytokine of VEGF.Methods:1. BM-MSCs are separated by gradient centrifugation from rabbit bone marrow and adherent culture on the common flask, then observing and testing the biologic characters of BM-MSCs induced by culturing with a conditional medium.2. Improve the parallel flow chamber device in order to better imitate hemodynamic fluid shear stress action on the endothelial cells, through changing the negative pressure into positive pressure in the device .
