The proliferation, differentiation and apoptosis of renal cells and the balance between the synthesis and degradation of extracellular matrix (ECM) in kidney play important roles in the development of kidney and renal disease. Tissue inhibitors of metalloproteinases (TIMPs) is the specific inhibitor of matrix metallopteinases (MMPs). TIMP-1 is a major member of TIMPs family. We already found that TIMP-1 could effect on the matebolism of ECM and the apoptosis of rat glomerular mesangial cells (RMC). But the mechanisms are still not clear.RNA interference (RNAi) is a natural mechanism by which small interfering RNA (siRNA) operates to specifically and potently downregulate the expression of a target gene. This downregulation has been through to predominantly function at the level of mRNA, as post-transcriptional gene silencing. The first part of this study is to investigate the effects of TIMP-1 shRNA (small hairpin RNA, shRNA) medicated by pEGFP-1 on the apoptosis of RMC and ECM in vitro.The messanger RNA (mRNA) of TIMP-1 was searched from Gene Bank.
