The clinical need for an alternative to donor corneal tissue has encouraged much interest in recent years. An artificial cornea must fulfill the functions of the cornea it replaces. More recently, the idea of a bio-engineered cornea has risen. Corneal equivalents have been reconstructed by tissue engineering method. They already have their potential uses in the biomedical world: as replacements for animals in toxicology testing and pharmacological studies, as well as in basic research into cell-cell and cell-matrix interactions of corneal wound healing. Is it possible to use an in vitro reconstructed tissue engineering cornea for corneal transplant? In this study, a novel corneal scaffold is going to be prepared and used to reconstruct tissue engineering cornea. 1. Preparation of porcine acellular corneal matrix (PACM)Fresh porcine corneas were decellularized by Triton combined with 0.25% trypsin; Triton combine with Dnase, Rnase; 0.25% trypsin combine with and Dna Rnase; Draw the materials in different times, the samples were observed by HE stain and light microscope; scanning electron microscope.Rabbit cornea stromal cells were seeded onto the PACM for 3-5days, and then...
