The most crucial immunological abnormality of bronchial asthma is imbalanced in the number and (or) the function between Th1 cells and Th2 cells, which performs Th2 cells' increased number and sthenic function while Th1 cells' decreased number and lower function. B7-CD28/CTLA-4 costimulatory path way is the most essential one in T cell activation, which is worthy to be studied in asthmatic research.Objective To explore the therapeutic function of anti-B7-1antibody in the mouse model of asthma.Methods Female BALB/c mice (weight 18-20g and 6-8 wk old) were divided into 4 group, group A: control-treated group (n=10), group B: asthmatic group (n=10), group C: IgG group (n=10), group D: B7-1 group (n=10). Mice in group B,C,D were sensitized to 10% OVA by intraperitoneal injections in first day, then they were exposed to 5% OVA aerosol challenges for 30 min on consecutive seven days after two weeks, and they were respectively treated to saline,IgG,anti-B7-1antibody by intraperitoneal injections in 2nd,3rd,4th days and before one hour of challenges in 15th,17th days. Mice in group A were given saline as above methods. After one hour of last challenges all mice were killed to collect samples, measure the level of IL-4,INF-γand the number of kinds of cells in the bronchoalveolar lavage fluid (BALF) and observe pathological changes of mice lungs.Results (1) The symptom and physical sign were more obvious in group B,C than in group A, while mice in group A had no asthmatic onset. (2) In group D the level of IL-4 was descended and INF-γwas ascended in BALF compared with group B,C, but it was reverse compared with group A, and both of differences were statistically significant (p<0.05). (3) In group D the number of total cells and eosinophils (EOS) in BALF was decreased compared with group B,C, but it was also reverse compared with group A, and differences between them were statistically significant (p<0.05).
