[Objective] We generated transgenic (Tg/+) mice that overexpress a heat shock protein 27 (Hsp27) and then to determine the effects of heat shock protein 27 on the isoproterenol(ISO)-induced cardiac hypertrophy in early period.[Methods] A cDNA encoding human heat shock protein 27 was subcloned into SalI-HindIII sites of pBS II-SK+ containing the a -MHC promoter,BamHI-digested linear transgene consistent with the a -MHC gene promoter, Hsp27 cDNA, and poly(A) of the human growth hormone (HGH) was microinjected into pronuclei of fertilized eggs from CBA mice. Isoproterenol was administered intraperitoneally once-daily with 30mg · kg-1d-1 for 7 days. The control mice were injected with physiological saline in same regimen with isoproterenol.Then to determine the protection effects of overexpression of heat shock protein 27 on the isoproterenol-induced cardiac hypertrophy in early period.
