Objective To research and discuss the relativity between immune function change of mouse with HCMV innate latent infection and Alzheimer's Disease -like Pathological Change of Brain of mouse.Methods Three groups mouse with innate HCMV latent infection (A group), HCMV reactivation infection (B group) or HF cell control (C group) were selected randomly, every group includes 6 mouses aged 18-20 months. After mouses are killed by bleeding from mouse eyepit, peripheral blood and brain assemble spleen of mouses were tested by following technical: (1) How HCMV infection estate of these mouse was validated by virus separation in base of cells co-cultivation of HF and mouse brain cells cultures, gene testing through PCR and RT-PCR assembled hybridization in situ. (2) Test of pathological and molecular change about Alzheimer's disease included following: Hematoxylin-eosin (H.E.) stain tested construction, pathological change and biological characteristic of mouse brain tissue ; Congo red stain tested amyloid-like plaque correlated AD; Silver stain tested neural fibre tangle related AD; (4) Expression quantity of amyloid- β precursor protein (APP) was observed by competitive RT-PCR; (3) T lymphocyte subgroups including CD-3+/CD4+/CD8+ molecule in peripheral blood and spleen cells wre tested by flow cell meter. (4) HE staining of spleen tissue was undertaken in order to observe change of lymphocytes quality and tissue construction in spleen.(5) T lymphocyte subgroups including CD3+/CD4+/CD8+ molecule in spleen cells were tested by flow cell meter. |
