| AIM:The acute, chronic inflammatory and immune inflammatory (adjuvant arthritis, AA) models were established in order to investigate the anti-inflammatory and immunomodulatory effects of hesperidin and its mechanism, meanwhile a series of immumopharmacological experiments in vivo and in vitro were adopted. METHODS AND RESULTS:1. Anti-inflammatory actions of HDNFour inflammatory models were used to investigate the anti-inflammatory actions of HDN, which were dimethylbenzene-induced mouse ear edema, carrageenan-induced rat hind paws swelling, cotton-induced mouse granuloma and FCA-induced AA models.HDN could remarkably inhibit the ear edema induced by dimethylbenzene in mice at the dose of 120,240 mg·kg-1, and reduce the swelling of rats hind paws induced by carrageenan, the weight of granuloma induced by cotton in rats, primary inflammatory reaction in AA rats at the dose of 40,80,160 mg-kg"1 respectively. The results showed HDN have a remarkable anti-inflammatory action.2. Immunoregulatory actions of HDNThe immunosuppressed mice were induced by cyclophosphamide (Cy, ip.). Index of immune organs was weighed and calculated. Phagocytosis of mononuclear macrophage was determined by cleaning carbon particle method. Spectrophotography was used to estimate levels of serum hemolysis IgG, IgM. Plaque forming cell were determined with QHS. Splenocyte lymphocytes proliferation was measured by MTT assay. The mouse DTH model induced by DNFB was used to study the effects of hesperidin on the levels of DTH and subset of T lymphocyte. |
