Objective To study the effects of endostatin on the growth and metastasis of breastcancer.Methods Model simulating human breast cancer was established by hypodermicimplantation of histologically intact tumor tissue into nude mice. Endostatin wasadministered sc at dose of 20.0mg/kg,10.0mg/kg and Omg/kg every other day forseven weeks. Eight weeks after implantation, tumor size, inhibition rate, intratumoralmicrovessel density, apoptotic index, and the presence of metastasis were evaluatedrespectively after the mice were sacrificed.Result Compared with the untreated control, growth of the tumor was significantlyreduced in size in mice treated with endostatin with an inhibition of rate91.3%,58.4%,0 at the dosage of 20.0mg/kg,10.0mg/kg and Omg/kg, respectively. TheMVD was also decreased significantly in the treated mice [(1.8±0.4) versus(4.6±0.9 ) ,(9.6±1.2 ) ]. The AI was increased significantly in the treatedmice[(12.4±4.9) % versus (8.3±2.6) %,(3.2±0.8) %]. The growth of breast cancerimplanted in nude mice were significantly inhibited (P<0.05) .The incidences ofliver (lung) was also significantly inhibited in the treated mice at dose of 20mg/kg (P<0.05).Conclusions Endostation can inhibit growth and metastasis of human breast cancerimplanted in nude mice effectively by inhibiting tumor angiogenesis and inducingapoptosis in breast cancer. And its effect is a dose-dependent manner.
