Mechanism Of The Signal Pathway Of ALR Promoting Liver Regeneration

Objective The promotion of ALR to liver regeneration has been verified by animal experiment. But if ALR can stimulate hepatocyte proliferation in vitro directly is still in controversy. The relationship of ALR and other hepatic cytokines is unclear in the progress of liver regeneration. The purpose is to investigate the mechanisms of the signal pathway of ALR promoting liver regeneration studied by animal and cell experiment.Materials and methods 1 .The promotion of ALR to liver regeneration after PH was evaluated by rate of liver regeneration. 2.These experiments were performed by MTT including the effects of ALR and animal serum of liver regeneration on damaged hepatocytes proliferation, ALR on hepatocytes proliferation inhibited by TGF-β₁, KCCM+/ICCM+ on damaged hepatocytes proliferation and ALR on Ito proliferation. 3.The trend of ALR expression in liver after PH, ALR binding to KC membrane and effect of ALR on expression of IL-6, TGF-β₁ in KC and TGF-α, α-SMA in Ito cell were determined by immunohistochemistry. 4. The concentration of TGF-a in ICCM+ was detected by radioimmunoassay. 5. Effect of ALR on expression of TGF-β₁ in Ito cell and expression of ALR in hepatocyte were examined by Western blot and immunohistochemistry.Results 1. ALR could promote liver regeneration after PH and ALR with animal serum of liver regeneration could stimulate proliferation of damaged hepatocytes. 2. In liver, immunostaining positive signal of ALR was decreased in the 2nd and 3rd day after PH, and then it was increased to the normal level in the 6th day after PH with the termination of liver regeneration progress. 3. ALR could antagonize the inhibiting role of TGF-β₁ on hepatocyte proliferation. 4. Expression of ALR in hepatocyte was increased after administration of ALR. 5.
