This thesis makes up of two primary parts: first, the separation and analysis of active ingredients from traditional Chinese medicines (TCM) by capillary electrophoresis, second, the interaction analysis between single drug and receptor by capillary electrophoresis.A capillary zone electrophoresis (CZE) was developed for the separation of Alkaloid compounds from Coptis chinensis. Analysis was performed on a polyimide-coated fused silica capillary. The detection wavelength was 254 run. According to the experimental results, the optimum separation condition was selected as fallows: 60 mmol·L-1 phosphate and 40% ethanol (v/v), pH 7.0, as buffer solution, and 22 kV as applied voltage. Under this experimental condition, the extracts were separated satisfactorily. This method could be applied to the ingredient analysis and further quality control of Coptis chinensis and its medicinal preparations.The work is to introduce high performance capillary electrophoresis method (HPCE) for the separation of Berberine extracted from Fufang Xialian Capsule.The buffer solution employed was 60 mmol·L-1 phosphate and 35% methanol. The capillary used was 50 μm in i.d., 50 cmin effective length, 60 cm in total length. UV detection wavelength was 265 run. The voltage was 14 kV. Under this experimental condition, the extracts were separated to three active ingredients (or active sites) in 12 min. The developed method could be applied for the ingredient analysis and quality control of Fufang Xialian Capsule.Human serum albumin (HSA) was selected as general receptor, and the interaction between HSA and the main alkaloids of Coptis chinesnsis and Ephedra sinica was studied using HPCE/FA, the binding constants of Berberin, Palmatine and Jatrorrhizine with HSA were k=1.09×105, k=1.41×105, k=1.88×105 respectively. The results show that the combination of three types of alkaloid with HSA is mainly static and hydrophobic action, and hydrophobic action is the key. The binding constants of Ephedrine Hydrochloride and Pseudoephedirne Hydrochloride with HSA were k= 1.83×104 , k= 1.96 ×104 respectively, which shows that the space configuration of these types of alkaloids have a great influence on binding constants. The detection of these binding parameters can provide important information for the transportation, distribution and activity of alkaloid substance in human bodies, and lay a foundation from the interaction between this drug and biological receptor moleculeDeficient in stern detached quality criteria and reliable scientific data to verify its effectiveness and security, TCM is still out of the main international medicine market. Combining the two strong functions of CE, that is to say, interaction analysis andingredients separation, the simultaneous studies of pharmacodynamics and active principles of TCM could be realized, which will help to broaden application range of CE, and to accelerate modernization of traditional Chinese medicines.
