Objective:To detect the expression lever of PTEN,cell cycle control protein P27and Ki-67 as well as the deletion of PTEN gene in malignant melanoma(MM) and melanocytic nevi. And to discuss the biological value of proteins,PTEN,P27,Ki-67 in malignant melanoma and the relationship among them,to investigate the effect of the proteins on the transformation from melanocytic nevi to malignant meianoma.Then to explore the relationship between cell proliferation and cell apoptosis in the occurrence and development of malignant melanoma.Methods:Immunohistochemistry was performed in 20 melanocytic nevi and 21 malignant melanoma to evaluate the alterations of the PTEN,P27,Ki-67 proteins. Polymerase chain reaction (PCR) was performed in 10 melanocytic nevus and 10 malignant melanoma to detect the loss rate of PTEN gene.Results:1.PTEN protein:The positive expression rate was 100%(20/20) in melanocytic nevi and 33.3% in malignant melanoma.
