The Pathologic Study Of Label-Retaining Cell In Gastric Poorly Differentiated Adenocacinoma

[Objective]Now in the world many theories argue there is a small fraction of cells in the tumor , called "tumor initiating cell" or "cancer stem cell" .This subpopulation cells in the tumor are tumorigenic cells, which elucidate the pathway that may account for their tumorigenic potential , its heterogeneity and specificity maybe decide the heterogeneity and specificity of the whole tumor mass. This small fraction cell has close relation with development and drug resistance of the tumor. Now cancer stem cell theory could be a shining light on the research horizon. But whether is really there such this small population of tumorigenic cells in tumors? What like is its morphology and biology? Whether has it definit markers? The purpose of this research is to use the specificity of tumor initiating cell—long cell cycle, which can abstract 5—bromodeoxyuridine (BrdU), which is thymidine analogue and label-retaining cell(LRC) can retain BrdU for so long time than other cells. Then search for such cell by immunohistochemistry and research its morphologic specificity .This study hope that it can provide new methods and new thoughts for cancer stem cell theory.[Method]1. Gastric adenocarcinoma model establishment in nude mice: culture the gastric adenocarcinoma cell line BGC-823 and implant them to the right oxter of the nude mice , after three days the oxter of some nude mice can detect tumor mass.2. After ten days all nude mice can be detected obvious tumor mass. Then BrdU 2mg inject into peritoneum of nude mice, every other day for one week.
