| OBJECTIVE: To investigate the effect of N3-o-toluyl- Flulorouracil (TFu) on FRH-0201 cell and to test new drugs for the chemotherapy of cholangiocarcinoma. METHODS: Morphological changes were observed by light microscopy and fluorescence microscopy. The effect of TFu on FRH-0201 cell was examined by clone formation. Cell cycle and apoptosis were examined by flow-cytometric analysis. DNA ladder was used to detect apoptosis. To compare the drug susceptibility of TFu with 5-Fu and several other common drugs by MTT colorimetric assay.RESULTS: It appears cytotoxicity when the concentration of TFu is >4mmol/L and no significant morphological changes when <0.016mmol/L. TFu significantly inhibited the proliferation and clone formation of FRH-0201 cell line in vitro within concentration of 0.0164mmol/L (P< 0.05). Morphological character of apoptosis was observed. The inhibition ratio was dependent on arsenic concentration and time. The inhibition ratio of TFu is higher ( P< 0.05) than 5-Fu with the same concentration of 0.016lmmol/L and same time of 72h. There is no remarkable difference when the concentration is 4mmol/L or the time is shorter or longer than 72h ( P> 0.05). The Flow cytometric analysis indicated that at 24h and 48h the percentage of G1 phase with TFu (83.18%,94.49%) and 5-Fu (61.82%,92.61%) increased remarkably compared with the control group(52.32%,35.96%) and the percentage of S phase with TFu(16.82%,5.51%) and with 5-Fu(38.18%,7.39%) decreased remarkably compared with the control group(64.04%,47.68%); The apoptosis ratio at 48 and 72h induced by TFu(12.22%,44.57%) and 5-Fu(4.67%,12.56%) increased remarkably compared... |
