ObectiveTo investigate the expression of HIF-1α,VEGF and ultrastructural changes in the retina of diabetic rats,and to evaluate the role of them in the pathogenesis of DR. It may provide the experiment evidence for diagnosis and prevention in the early stage of DR.Methods132 SD rats were randomly divided into two groups: control group and DR group. Diabetes was induced by STZ intraperitoneal injection in DR group. Retinal paraffin sections were stained by immunohistochemical (SP) method to investigate the changes of the activity of HIF-1αand the expression of VEGF, the reverse transcription polymerase chain reaction (RT-PCR) method was used to examine the HIF-1α,VEGFmRNA expression level in retina respectively at 1, 3, 6 months after STZ injection. meanwhile, transmission electron microscope method was used to observe the ultrastructure of retina in three stage of DR.Results1,The result of immunohistochemistry demonstrated that HIF-1α,VEGF were not expressed in control group.
