Objective: To study the expression of 5-lipoxygenase(5-LOX) in gastric cancer and normal cancerous beside tissue and investigate the relationship between the expression of 5-LOX and clinical pathological factors;To observe the effects of AA861,a specific 5-lipoxygenase inhibitor, on cell growth and the expression of 5-LOX gene in gastric cancer cell line AGS.Methods: the expression of 5-LOX mRNA in 30 cases of human gastric cancer and 30 cases of normal cancerous beside tissue were analyzed by reverse transcription polymerase chain reaction (RT-PCR).the expression of 5-LOX protein in 40 cases of human gastric cancer and 30 cases of normal cancerous beside tissue were examined by immunohistochemistry technique, and to investigate the relationship between the expression of 5-LOX and clinical pathological factors.Gastric cancer cell lines AGS were co-cultured with AA861.Cell growth was surveyed by cell count and MTT assay. Apoptosis-related changes in morphology were observed by transmission electron microscopy (TEM).Apoptotic index(AI) was counted by TDT-mediated dUPT -biotin nick end-labeling (TUNEL) and flow cytometric analysis assay(FCM).the effect of AA861 on the expression of 5-LOX was tested by semi-RT-PCR and immunocytochemistry.Results: The expression of 5-LOX mRNA and protein were detected in gastric cancer tissue.The positive rates of 5-LOX mRNA and protein expression in gastric cancer were 76.7%,72.5%,and in normal cancerous beside tissue were 17%,23%.Expression of 5-LOX mRNA and protein in gastric cancer were significant higher than in normal cancerous beside tissue (P<0.05).
