| Objective:To study the expression of Skp2 protein in colorectal carcinoma and its relationship with the expression of p27kip1.To explore its role in the tumorigenesis and development in colorectal carcinoma. Methods:Immunohistochemical staining of Skp2 and p27kip1 were performed by the standard streptavidin-peroxidate (SP)technique for the paraffin-embeded sections of 80 colorectal carcinomas ,20 colorectal adenomas,20 colorectal polypi, 20 normal colorectal tissues. A computerized image analysis system was used to analyse the expression of Skp2 and p27kip1 . The relationship between expression of Skp2 with the clinicopathologic parameters and the expression of p27kip1 was analyzed respectively. Results:1. The Skp2 protein expression was mainly found in nucleolus. The positiverate in carcinoma , adenomas and normal tissues were 93.75 %( 75/80), 55 %( 11/20) and 0 %(0/20)respectively; the positive rate of Skp2 was higher than those in adenomas and normal tissues (P<0.001). The expression of Skp2 was significantly associated with the loss of tumor differentiation and lymph node metastasis ( P< 0. 01) , but not with patients age,sex,tumor location and deep invasion( P > 0. 05 ).2. The expressions of Skp2 gradually increased in the order of normal mucosa, adenoma to colorectal cancer ( P < 0. 05 ) , while the expression of p27kip1gradually decreased in the same order ( P < 0. 05).
