Ginsenoside Rbl and Rgl Can Attenuate Tau Protein Hyperphosphorylation and increase Tubulin acetylation in GSK3β-overexpressing SH-SY5Y cell lineObjective construct GSK3β-overexpressing SH-SY5Y cell model systems and observe the expression of phosphorylating tau with serl99/ser/202, thr231,thr205 residues and the extent of tubulin acetylation,then explore the protective effect of Gensenoside Rg1 and Rb1 on the abnormal phosphorylation of tau and the extent of tubulin acetylation of this cell model.Methods The cDNA of full-length GSK3β construct was subcloned into the mammalian expression vector,pbudce4.1. The integrity of the GSK3P construct was confirmed by sequence analysis. GSK3P was transiently transfected into SH-SY5Y cells using Lipofectamine2000 transfection reagent. The expression of GSK3β and phosphorylating GSK3β were measured by wesern blot. On the GSK3β-overexpressing condition, the expression of phosphorylating tau with ser199/ser/202, thr231 ,thr205 residues and total tau and the extent of tubulin acetylation were observed by wesern blot. Meanwhile, the cell mode was administered different doses of Gensenoside Rg1 and Rbl (Rg1: 20μmol/L, Rb1: 10μmol/L)for twelve or twenty-four consecutive hours after transfection.Results 1.Thirty-six hours After transfection, The expression of GSK3β and phosphorylating GSK3β in SH-SY5Y cell model systems were significantly increased compared with normal groups and vector groups(p<0.05), after fourty-eight hours,the level of phosphorylating tau with ser199/ser/202, thr231,thr205 residues were obviously increased,but total tau was changed little.
