| Objective: To investigate the effect of somatostatin analogue Octreotide on the expression of Hpa in gastric cancer cell SGC-7901 in vitro and vivo.Methods:(1)The SGC-7901 human gastric cancer cells in logarithmic growth period were made into single cell suspension , and then co-cultured with different concentrations of Octreotide for 24 and 48 hours. The expression of Hpa was detected by semiquantitative RT-PCR and immunocytochemical method respectively. (2)A nude mouse model of xenotransplanted human gastric cancer was established by implanting SGC-7901 cells under the skin of nude mice. And then the mice were randomly divided into four groups and treated with Octreotide, 5-FU, Octreotide plus 5-FU or nomal saline for six weeks respectively. The expression of heparanase and bFGF were examined by immunohistochemical technique.Results: (1)After co-culturing for 24 hours, the expression level of HpamRNA in Octreotide groups of different concentrations and control group were 0.65±0.13(t=-10.72, P=0.000, vs control), 1.05±0.08(t=-4.98, P=0.001, vs control), 1.36±0.07(t=-0.43, P=0.68, vs control), 1.48±0.08(t=1.20, P=0.26, vs control), 1.39±0.04 respectively; After co-culturing for 48 hours, the expression level of HpamRNA in Octreotide groups of different concentrations and control group were 0.12±0.03(t=-15.52, P=0.000, vs control), 0.55±0.10(t=-9.76, P=0.000, vs control), 1.23±0.07(t=-0.71, P=0.49, vs control), 1.21±0.15(t=-0.98, P=0.35, vs control), 1.28±0.06 respectively. There was significantly difference between trial groups and control group in the expression level of Hpa protein, after gastric cancer cells co-cultured with Octreotide of 1×10-5mol/L and 1×10-6mol/L for 24 hours and 48 hours respectively (F=94.346,P<0.01). (2)In the tumor tissues, the weakly positive expression rates of Hpa in Octreotide group,5-FU group,combined group and control group were 25.0%,41.7%,25.0% and 16.7% respectively; The strongly positive expression rates were 16.7%,33.3%,8.3% and 75.0% respectively.
