Objective:To investigate the significance of relationship between the expression of chemokine receptor CXCR4 and CXCR2 in the tissue of colorectal carcinomas and evaluate its role in invasion and metastasis in colorectal carcinomas.Methods:The levels of chemokine receptors CXCR4 and CXCR2 in the tissue were detected by immunohistochemical Elivision plus method and RT-PCR method on protein and mRNA levels in 46 cases of colorectal carcinoma and 30 cases of benign colorectal tumor and 30 cases of normal colorectal tissue. The differences were compared between benign and malignant diseases, clinical stages.Results:1. By immunohistochemical Elivision plus method,the positive expression rates of CXCR4 in colorectal carcinomas were 52.3%, the positive expression rates of CXCR4 in benign colorectal tumor and normal colorectal tissue were 25.0% and 13.3%.By RT-PCR method, the positive expression rates of CXCR4 in colorectal carcinomas were 67.4%, the positive expression rates of CXCR4 in benign colorectal tumor and normal colorectal tissu were 25.0% and 20.0%.2. By immunohistochemical Elivision plus method, the positive expression rates of CXCR2 in colorectal carcinomas were 69.6%, the positive expression rates of CXCR2 in benign colorectal tumor and normal colorectal tissue were 35.0% and 33.3%. By RT-PCR method, the positive expression rates of CXCR2 in colorectal carcinomas were 69.6%, the positive expression rates of CXCR2 in benign colorectal tumor and normal colorectal tissue were45.0% and 33.3 %.
