Brucella is a facultative intracellular gram-negative bacterium that is able to infect man and a broad range of wildlife domestic mammals, and is also a focus of concern as potential biological warfare agent. The zoonotic disease and huge economic losses caused by brucella had been the focus all over the world.All the classical serological procedure being in used in diagnosis of brucellosis, such as complement fixation test, rose bengal plate test, sero-agglutination test and enzyme-linked immunosorbent assay, are mainly based on the detection of antibody directed against the lipopolysaccharide (LPS) portion of the cell membrane. This dominant antigen is common to both attenuated vaccinal strain and virulent brucella strain. Therefore distinguish between the result of infection and vaccination is difficult. In addition, test based on anti-LPS antibodies have extensive cross-reaction with other gram-negative bacteria, which further impaired the unambiguous serologic diagnosis of brucellosis. Among different approach, the development of test based on antibodies directed against protein antigen of brucella could help to solve the problems.The live attenuated strain is considered to be the best vaccine of brucellosis.
