As traditional Chinese medicines, Sophra flavescens Ait. and sophora alopecuraides L. have been used for the treatment of many diseases for thousands of years. Its alkaloid extract, marine, has long been extensively used in China. Marine is a mix alkaloid, which consists of more than 98% oxymatrine (OM) and a little oxysophocarpine. It is reported that oxymatrine has a lot of pharmacological functions. Basic and clinical researches suggested that OM had the following pharmacological effects such as anti-virus, protecting hepatocytes, anti-hepatic fibrosis, immune regulation, etc. In clinical therapy OM possesses beneficial effect on chronic hepatitis B virus (HBV) infection.On the other hand, three times a day administration is inconvenient to raise patents' compliance. The purpose of the present study was designed to develop a kind of Marine sustained release pellets with 1.0mm diameter which would decrease adverse effects and improve its bioavailability, and which would be more convenient for patients to use.RP-HPLC was utilized to determine the OM content in pellets, and assay release of OM sustained release pellets.The preformulation results showed that Marine is easily soluble in water and various physiological pH fluids and the n-octanol/water partition coefficients increase with pH rising in various physiological pH fluids(Papp:0.160.38).Marine pellets were prepared by means of power layering with the centrifugal granulation equipment.
