| Sargassum Pallidum(Tum.)C.Ag,which is accepted as Chinese traditional medicine(CTM)in the China Herbal,has attracted much attention for its multiple biological activities.Studies have been carried out to investigate the potential anti-tumor compounds derived from marine alga,Sargassum Pallidum(Tum.)C.Ag.Fucoidans from the Traditional Chinese Medicine Algae Sargassum Pallidum (Tum.)C.Ag.were prepared by different methods for extraction and purification,andthe composition and structure of the polysaccharides were elucidated.Crude fucoidans C1 and C2 were obtained by acid method and aqueous method respectively.C1 was purified by fractional precipitation with alcohol and calcium chloride to get F1 and F2.C2 was purified to get F3 and F4.The result showed that crude polysaccharides of acid extraction were suitable to be purified by fractional precipitation and crude polysaccharides of aqueous extraction was suitable to be , purified by calcium chloride method.P1,P2 and P3 were obtained by anion exchange column(Q-Sepharose Fast Flow)and chromatography column(Sephadex G-200) from F4 successively.Chemical components,molecular weight,monosaccharide compositions,periodate oxidation analysis,methylation and IR confirmed the characteristics ofPl,P2 and P3.P1 contained 4.78%sulfate;molecular weight is 494.4KDa;monosaccharide composition are fucose,xylose,mannose,glucose and galactose with a molar percentage ratio of 43.4:33.3:6.2:4.3:12.8.P2 contained 1 5.34% sulfate;molecular weight is 61.5KDa;monosaccharide composition is fucose,xylose,mannose,galactose,glucose uronic acid and mannose uronic acid with a ratio of 27.8:12.0:14.9:12.5:29.0:3.8;the linkage is:Thefucoidan constituent is mainly composed of l→3-linked fucose,mannose units 1→4-1inked xylose units and l→3,4-1inked mannose.P3 contained 18.71% sulfate;molecular weight is 167.6KDa;monosaccharide composition is fucose,xylose,galactose and glucose uronic acid with a ratio of 49.4: 2.4:22.6:25.6;the linkage is:The fucoidan constituent is mainly composed of 1→3-1inked fucose,galactose units,1→4-linked xylose units and 1→2,3-1inked galactose. |
