Ulva pertusa is an edible macroalgae of Chlorophyta, and has many traditional active properties such as against heatstroke, hydropsy and so on. In this paper, several polysaccharides from Ulva pertusa are separated using different kinds of extraction and purification methods. The chemical characterization and primary structure are determined by chemical and modern instruments analysis techniques. The dry algal powder was hydrolyzed respectively by HCl and H2SO4 .The total carbohydrate content was determined respectively by the phenol-sulfuric method and anthrone-sulfuric method. The result showed that the total carbohydrate content is about 35%.The algal flour was extracted sequentially with cool water, hot water and hot 1 mol/L NaOH solution at 100℃. Three types of polysaccharides were obtained, and respectively named as UCPS, UWPS and UAPS. The chemical characterization of UCPS and UWPS are defined. The results showed that the UCW mainly contained rhamnose, xylose, glucose and little galactose, UWPS contained rhamnose, xylose and glucose. The carbohydrate contents of UCPS and UWPS are 55.37% and 63.46%, protein contents 6.35% and 4.05%, sulfate ester contents 24.99% and 17.88%, uronic acid contents 32.09% and 22.62%. The relative molecular weight 324kD and 310kD.The polysaccharides from UCPS and UWPS were fractionated by anion exchange chromatography on Q Sepharose Fast Flow.
