[Objective]This study was designed to explore the effect of Salvia yunnanensis C.H.Wright on the fibroblast of scar and the pig back scar model about its biological behavior and its pathological changes. Reveal the mechanism to provide basic theory for clinical precaution and treatment to scars at molecular and pathological level. [Methods] 1. The fibroblasts of scars were cultivated in culture fluid containing different concentration Salvia yunnanensis C.H.Wright(0.1g/ml, 0.2g/ml, 0.4g/ml, 0.6g/ml) . The results were observed and analyzed by light microscope, MTT assay, transmission electron microscope and flow cytometry. And flow cytometry analyzed cell cycle and TUNNEL and the content of DNA, Bcl-2 and Fas. 2. Hypertrophic scar models were made on pig back in vivo by different concentration Salvia yunnanensis C.H.Wright injection, detected by HE staining, Masson's staining, transmission electron microscope and in situ hybridization. Using these experimental analyse, to study the biological behavior and pathological changes of hypertrophic scar fibroblasts affected by Salvia yunnanensis C.H.Wright.[Results] 1. In vitro: compared with the control group, by LM, the density of cells depressed and the spindle-shape cell were discovered becoming retractation and abnormity. MTT assays showed significant difference between the two groups (P<0. 05). By FCM: compared with control groups, percentages of the cells in the growth phases( S-G2-M) of cell cycle decreased and the level of apoptosis increased in the experimental groups and higher expression quantities in Fas and lower in Bcl-2 . 2. In vito (pig back scar model): Compared with the control group, by pathological staining, the experiment group were less fibroblasts and the collagen density reduced and arranged in regulation. The spaces between fiber bundles were increased. By TEM, compared with the control group, there were less fibroblast than fibro and undeveloped rough endoplasmic reticulum. Heterochromatins were very abundance and an arrangement of collagen fibers which were orderly.
