Objective: Milrinone , phosphodiesterase III (PDE-III) inhibitor,is a new vasoactive agent. A lot of research has shown that Milrinone has a beneficial action in improving cardiovascular function, and can improve the aerobic metabolism of system, while the Effect of Milrinone on Oxygen Free Radical of shock have not been reported at present.In this research it investigates the Effect of Milrinone on Oxygen Free Radical of shock by testing arterial malonaldehyde(MDA) of rabbits in the superior mesenteric artery occlusion (SMAO) shock. Methods: Sixteen healthy rabbits were duplicated SMAO model, then randomly divided them into two groups(n=8): group A was control group ,for continuously being pumped normal saline 0.1ml.kg-1.mim-1 by jugular vein during the time of shock; group B was treated group,for being intravenous injected slowly Milrinone 50 u g.kg-1 (in 10 minutes) followed by 0.5 u g. kg-1.min-1 was administered. In the course the experiment,heart rate(HR), Mean arterial blood pressure(MAP), and SMAF were recorded, and arterial blood MDA, carotid and superior mesenteric venous blood gas analysis were detected before shock(-lh) and after shock(0h,1h,2h).The oxygen delivery(DO2), oxygen consumption(VO2) and oxygen uptaking rate(O2ER) were calculated by formula. Results:Compared with before shock(-1h), the MAP,HR and SMAF of A and B groups were significantly decreased after removed artery clamp and shock (Oh), there was a significant difference in MAP,HR and SMAF (P<0.05 or P<0.01).
